Officials said the a tire was kicked up and hit the motorcyclist in the head at mile marker 67 at about 7 a.m.
The motorcycle rider was then thrown from the bike and ran over by a truck, according to the Florida Highway Patrol.
FHP said the truck, believed to be a dark grey, heavy-duty F250 or F350 with damage, fled the scene.
Delays on I-4 are expected for several hours as officials clean up and investigate.
"Due to the gruesome nature of this crash, we cannot let drivers travel through the scene right now because there is not a lane that we can open that's not affected," said FHP Sgt. Kim Montes.
Drivers can enter I-4 at State Road 536 and miss the closure, or use International Drive or State Road 535 can be used as alternates.
